Mit dem Cloud-Aerosol Transport System (CATS) Experiment erfolgreich auf einer Plattform außerhalb des japanischen Kibō-Moduls installiert verbrachte die Expedition 42-Mannschaft den Freitag arbeitend mit Biowissenschaften, Verbrennung und einer Vielzahl von anderen Experimenten. Das Combustion Integrated Rack (CIR), befindlich innerhalb des „Destiny“-Labormoduls, benötigt Treibstoff, so können Wissenschaftler Materialien entzünden, um das Verhalten von Flammen und Rauch im Weltraum zu studieren. Hardwareteile, welche Treibstoff speichern und liefern, einschließlich Zündungsspitzen, wurden am Freitag von Kommandant Barry Wilmore  (USA/NASA) auf dem CIR ersetzt, um die Haltevorrichtung im Betriebszustand zu halten. Terry Virts – mit Unterstützung von Samantha Cristoforetti und Ärzten am Boden – nahm teil an weiteren Augenprüfungen und unterzog sich Ultraschalluntersuchungen und einer Echokardiografie,  um mehr Informationen zu sammeln. Die Ocular Health Studie versucht, zu verstehen, wie Schwerelosigkeit die Augen von Besatzungsmitgliedern beeinflusst und zu erklären, warum einige Astronauten bei ihren Einsätzen im Raum über Sehstörungen berichten. Virts überprüfte auch Proben und transferierte Daten, die gesammelt wurden für das Coarsening in Solid Liquid Mixtures-2 (CSLM-2) Experiment, das möglichen Nutzen für Verbraucher- und Industrieprodukte bietet. Cristoforetti überprüfte die Fruchtfliegen an Bord der Internationalen Raumstation (ISS) und arbeitete später an der Wartung der Magvector-Studie, die beobachtet, wie das Erdmagnetfeld mit einem elektrischen Leiter interagiert.

Für Anfang nächsten Jahres sind drei US-Weltraumspaziergänge geplant und ISSBesatzungsmitglieder Kommandant Barry Wilmore (USA/NASA) und Bordingenieur Terry Virts bereiten Raumanzüge und Werkzeuge für die Außenbordeinsätze vor. Wilmore tauschte an einem Paar Raumanzügen Sekundärsauerstoffpacks aus, während Virts das Drehmoment an einem Pistolengriff-Werkzeug überprüfte. Nach den Arbeiten an den Raumanzügen absolvierte Wilmore mit Astronautin Samantha Cristoforetti RobotikTraining innerhalb der Cupola. Wilmore wird den Canadarm2-Roboterarm bedienen, um den SpaceXDragonRaumfrachter einzufangen, wenn dieser am Sonntagmorgen ankommt. Samantha wird den Kommandanten unterstützen während der Annäherung und des Rendezvous des kommerziellen Raumfrachters. Virts installierte auch eine Mittellinien-Kopplungs-Kamera zur Unterstützung des Andockens zwischen Dragon und dem “Harmony”-Verbindungsknoten. Dragon ist fällig für den Start im Rahmen der SpaceX CRS-5-Mission am Freitag um 1:20 p.m. EST und das Einfangen ist für Sonntag 6 a.m. geplant.  Die drei Astronauten arbeiteten auch an einer Vielzahl von Forschungen und studierten Verbrennungen und Botanik und lernten sogar die Bedienung der Basketball-großen Satelliten, die  innerhalb der ISS schweben. Die drei Kosmonauten im russischen Segment der Station arbeiteten auch an ihrer Aufgabenliste von Wissenschaft und Wartung.
