Die drei Expedition 43-Besatzungsmitglieder waren am Dienstag wieder bei der Arbeit an laufender fortgeschrittener Mikrogravitations-Forschung, von der die Menschen auf der Erde und zukünftige Besatzungsmitglieder auf  langfristigen Weltraummissionen profitieren. Die Mannschaft der Internationalen Raumstation (ISS) ist auch bereit für die Ankunft neuer Besatzungsmitglieder und eines privaten Raumtransporters. Kommandant Terry Virts nahm an einem zweiten Tag von Sehtests teil, für die Ocular Health Studie.  Samantha Cristoforetti setzte mehr Durchläufe des TripleLux-B-Experiments fort, dieses Experiment studiert Zellmechanismen, die eine Beeinträchtigung der Immunfunktionen in der Mikrogravitation verursachen. Schließlich arbeitete der Kosmonaut Anton Schkaplerow  (Russland/Roskosmos) an einem Paar russischer Untersuchungen, die die Effekte des Erdmagnetfeldes auf die ISS und Strahlenbelastung auf ein simuliertes Besatzungsmitglied, oder Modell, studieren. Inzwischen sind Sojus TMA-16M-Kommandant Gennadi Padalka (Russland/Roskosmos) und die Ein-Jahres-Besatzungsmitglieder Scott Kelly (USA/NASA) und Michail Kornijenko (Russland/Roskosmos) in der Endphase ihrer Vorbereitungen vor dem Start am Freitagnachmittag, um sich  Expedition 43 anzuschliessen. Kelly und Kornijenko werden bis März 2016 im Weltraum bleiben. Padalka wird am 11. September zur Erde zurückkehren.

Die Expedition 42-Besatzungsmitglieder Barry Wilmore, Alexander Samokutjajew (Russland/Roskosmos) und Jelena Serowa (Russland/Roskosmos) sind etwa zwei Tage entfernt vor Beendigung ihres Aufenthalts an Bord der Internationalen Raumstation (ISS). Das Trio packt Ausrüstung und reinigt Mannschaftsunterkünfte, während sie sich auf die Abkopplung ihres Raumschiffs Sojus TMA-14M am Mittwoch um 6:44 p.m. EDT (23:44 MEZ) und die Landung in Kasachstan um  10:07 p.m. (3:07 MEZ) vorbereitet. Wilmore, der Expedition 42-Kommandant, übergab am Dienstag während einer Kommandowechsel-Zeremonie die Kontrolle über das Orbitallabor an Terry Virts, der der Kommandant von ISS-Expedition 43 ist. Mittlerweile wurden fortgeschrittene Schwerelosigkeitsforschung und Labor-Wartung an Bord der ISS fortgesetzt. Unter den Experimenten studierte die Besatzung Körpergröße und -form im Weltraum für Größenbestimmung der Raumanzüge und beobachteten Entzündung der Atemwege bei Astronauten. Ethernet-Kabel wurden ebenfalls installiert im „Harmony“-Modul, was zukünftigen kommerziellen Raumschiffen ermöglichen wird, mit einem Paar kommenden International Docking Adapters (IDA) zu kommunizieren.

Kommandant Barry Wilmore und Bordingenieur Terry Virts überprüften am Mittwoch Außenbordeinsatz-Prozeduren. Sie sollen den ersten von drei Weltraumspaziergängen planmäßig am 20. Februar starten. Ein CubeSatKleinsatellit, der im Januar an Bord des kommerziellen SpaceXDragonWeltraumfrachters zur Internationalen Raumstation (ISS) geliefert wurde, wird am Donnerstagmorgen außerhalb des  japanischen Kibō-Moduls ausgesetzt. Es werden verschiedene Subsysteme im Weltraum getestet und enthält auch ein Amateurfunk-Experiment. ESA-Flugkontrolleure untersuchen ein Signal einer Störung in einer Stromkette, die Batterieleistung für das ESA Automated Transfer Vehicle 5 (ATV-5: Georges Lemaître) bietet. Drei weitere Stromketten operieren normal innerhalb des ATV-5 und die sechsköpfige Expedition 42-Besatzung setzt ihre normalen Aktivitäten fort.

Nach einer Woche medizinischer Forschungsaktivitäten begannen die Bewohner der Internationalen Raumstation (ISS) die neue Woche mit Schwerpunkt auf  Würmern, Fruchtfliegen und Pflanzen. Die winzigen Organismen bieten Wissenschaftlern ein Modell für größere Organismen und wie Schwerelosigkeit sich auswirkt auf solche Dinge wie die Immunität, Muskeln und Knochen. Botanikforschung im Weltraum hilft Wissenschaftlern zu verstehen, wie Pflanzenzellen und Wurzeln sich entwickeln und möglicherweise zukünftige Besatzungen auf Langzeitmissionen und interplanetarischer Erforschung unterstützen. Es finden zahlreiche Pflanzenstudien statt auf der Station, die nicht nur zukünftige Weltraummissionen unterstützen, aber möglicherweise die Pflanzenproduktionstechniken auf der Erde verbessern. Die Expedition 42-Besatzungsmitglieder arbeiteten auch an Frachttransfers zum und vom kommerziellen SpaceXDragonRaumfrachter sowie dem Weltraumfrachter Progress M-25M. Eine Reihe von Routinewartungsarbeiten stand auf dem Programm einschließlich hochfliegenden Sanitärarbeiten, Batteriewiederaufladungen eines Weltraumanzugs und Wissenschafts-Hardware- Installationen.
