Das Sojus TMA-16MRaumschiff startete um 3:42 p.m. EDT (1:42 a.m. am 28. März Ortszeit/20:42 MEZ) vom Kosmodrom Baikonur in Kasachstan zur  Internationalen Raumstation (ISS). Scott Kelly (USA/NASA),  die Kosmonauten Sojus TMA-16M-Kommandant Gennadi Padalka (Russland/Roskosmos) und Michail Kornijenko  (Russland/Roskosmos) trafen um 9:33 p.m. an der ISS ein. Das Raumschiff dockte um 9:36 p.m. am „Poisk“-Modul, die Luken wurden um 11:33 p.m. geöffnet. Kelly, Padalka und Kornijenko wurden von den aktuellen ISS -Bewohnern Expedition 43-Kommandant Terry Virts, Anton Schkaplerow und Samantha Cristoforetti begrüßt. Kelly und Kornijenko werden bis März 2016 im Weltraum bleiben. Padalka wird am 11. September zur Erde zurückkehren. Virts, Schkaplerow und Cristoforetti trafen im November an Bord ihres Sojus TMA-15M-Raumschiffs  an der ISS ein und werden bis zum 14. Mai an Bord bleiben.
